Innovent Biologics and Spero Therapeutics have entered into an exclusive licence agreement for IBI355 (SP001), a Phase 2-ready third-generation Fc-silent anti-CD40L antibody, for the development and commercialisation of treatments for autoimmune diseases outside Greater China.

  • Mechanism of Action: IBI355 is a humanised Fc-silent IgG1 monoclonal antibody targeting CD40L, designed to interrupt immune signalling involved in chronic inflammation while avoiding B-cell depletion and platelet activation associated with earlier anti-CD40L antibodies.
  • Clinical Development: Spero plans to initiate a Phase 2 clinical trial in patients with IgG4-related disease in the second quarter of 2027, while Innovent plans to begin a Phase 2 trial in China for Sjögren’s disease by early 2027.
  • Licensing Terms: Spero receives exclusive worldwide rights outside Greater China to develop, manufacture, and commercialise IBI355, while Innovent retains rights in Greater China and is eligible to receive upfront, milestone, and royalty payments under the agreement.
